SD-OCThasbecomeanessentialtoolforevaluatingmacularpathology;howeverseveralaspectsofdatacollectionandanalysis a?ect the accuracy of retinal thickness measurements. Here we evaluated sampling density, scan centering, and axial length compensationasfactorsa?ectingtheaccuracyofmacularthicknessmaps.Forty-threepatientswithvariousretinalpathologies and113normalsubjectswereimagedusingCirrusHD-OCT.ReducedB-scandensitywasassociatedwithincreasedinterpolation errorinETDRSmacularthicknessplots.Correctingforindividualdi?erencesinaxiallengthrevealedmodesterrorsinretinal thicknessmaps,whilemorepronouncederrorswereobservedwhentheETDRSplotwasnotpositionedatthecenterofthefovea (whichcanoccurasaresultoferrant?xation).Cumulativeerrorcanexceedhundredsofmicrons,evenunder“idealobserver” conditions.Thispreventableerrorisparticularlyrelevantwhenattemptingtocomparemacularthicknessmapstonormative databasesormeasuringtheareaorvolumeofretinalfeatures. 1.Introduction result from poor signal quality of the SD-OCT image or the outright
