Epidermalkeratinocytesareusedasacellsourceforautologousandallogeniccelltransplanttherapyforskinburns.Thequestion addressedhereistodeterminewhetherthecultureprocessmayinducecellular,molecular,orgeneticalterationsthatmightincrease theriskofcellulartransformation.Keratinocytesfromfourdi?erenthumandonorswereinvestigatedformolecularandcellular parametersindicativeoftransformationstatus,including(i)karyotype,(ii)telomerelength,(iii)proliferationrate,(iv)epithelial- mesenchymaltransition,(v)anchorage-independentgrowthpotential,and(vi)tumorigenicityinnudemice.Resultsshowthat, despiteincreasedcellsurvivalinonekeratinocytestrain,noneoftheculturesdisplayedcharacteristicsofcelltransformations, implyingthatthecultureprotocoldoesnotgenerateartefactsleadingtotheselectionoftransformedcells.Weconcludethatthe currentprotocoldoesnotresultinanincreasedriskoftumorigenicityoftransplantedcells. 1.Introduction ity in cultured
