Recently,c-aminobutyricacid(GABA)aswellastheassociatedsyntheticenzymesandreceptorshavealsobeenidentifiedin tastecells.GABAreducestaste-evokedATPsecretionfromReceptorcellsandisconsideredtobeaninhibitorytransmitterin tastebuds.However,todate,theidentityofGABAergictastecellsandthespecificstimulusforGABAreleasearenotwell understood.Inthepresentstudy,weusedgenetically-engineeredChinesehamsterovary(CHO)cellsstablyco-expressing GABAB receptors andGaqo5proteinstomeasureGABAreleasefromisolatedtaste buds.Werecordedrobustresponses fromGABAbiosensorswhentheywerepositionedagainsttastebudsisolatedfrommousecircumvallatepapillaeandthe budsweredepolarizedwithKClorastimulatedwithanacid(sour)taste.Incontrast,amixtureofsweetandbittertaste 2+ stimulididnottriggerGABArelease.KCl-oracid-evokedGABAsecretionfromtastebudswasCa -dependent;removing fromthebathingmediumeliminatedGABAsecretion.Finally,weisolatedindividualtastecellstoidentifytheoriginof 2+ Ca GABAsecretion.GABAwasreleasedonlyfromPresynaptic(TypeIII)cellsandnotfromReceptor(TypeII)cells.Previously,we reportedthat5-HTreleasedfromPresynapticcellsinhibitstaste-evokedATPsecretion.Combinedwiththerecentfindings that GABA depresses taste-evoked ATP secretion [1], the present results indicate that GABA and 5-HT are inhibitory transmittersinmousetastebudsandbothlikelyplayanimportantroleinmodulatingtasteresponses.
