AgeCorrectioninDementia–MatchingtoaHealthy Brain JuergenDukart1,3*,MatthiasL.Schroeter1,2,3,KarstenMueller1,TheAlzheimer’sDiseaseNeuroimaging Initiative 1MaxPlanckInstituteforHumanCognitiveandBrainSciences,Leipzig,Germany,2DayClinicofCognitiveNeurology,UniversityofLeipzig,Leipzig,Germany,3LIFE- LeipzigResearchCenterforCivilizationDiseases,UniversityofLeipzig,Germany Abstract Inrecentresearch,manyunivariateandmultivariateapproacheshavebeenproposedtoimproveautomaticclassificationof variousdementiasyndromesusingimagingdata.Someofthesemethodsdonotprovidethepossibilitytointegratepossible confoundingvariableslikeageintothestatisticalevaluation.Asimilarproblemsometimesexistsinclinicalstudies,asitisnot always possible to match different clinical groups to each other in all confounding variables, like for example, early-onset (age,65years)andlate-onset(age$65)patientswithAlzheimer’sdisease(AD).Here,weproposeasimplemethodtocontrol forpossibleeffectsofconfoundingvariablessuchasagepriortostatisticalevaluationofmagneticresonanceimaging(MRI) datausingsupportvectormachineclassification(SVM)orvoxel-basedmorphometry(VBM).WecompareSVMresultsforthe classificationof80ADpatientsand79healthycontrol subjects basedonMRIdatawithandwithoutprioragecorrection. Additionally,wecompareVBMresultsforthecomparisonofthreedifferentgroupsofADpatientsdifferinginagewiththesame groupofcontrolsubjectsobtainedwithoutincludingageascovariate,withageascovariateorwithprioragecorrectionusing theproposedmethod.SVMclassificationusingtheproposedmethodresultedinhigherbetween-groupclassificationaccuracy comparedtouncorrecteddata.Further,applyingtheproposedagecorrectionsubstantiallyimprovedunivariatedetectionof disease-relatedgreymatteratrophyusingVBMinADpatientsdifferinginagefromcontrolsubjects.Theresultssuggestthatthe approachproposedinthisworkisgenerallysuitedtocontrolforconfoundingvariablessuchasageinSVMorVBManalyses.
