Carr,DavidC.Baulcombe* DepartmentofPlantSciences,UniversityofCambridge,Cambridge,UnitedKingdom Abstract Background: Argonaute (AGO) proteins bind to small-interfering (si)RNAs and micro (mi)RNAs to target RNA silencing againstviruses,transgenesandinregulationofmRNAs.PlantsencodemultipleAGOproteinsbut,inArabidopsis,onlyAGO1 isknowntohaveanantiviralrole. Methodology/PrincipalFindings:TouncovertherolesofspecificAGOsinlimitingvirusaccumulationweinoculatedturnip crinklevirus(TCV)toArabidopsisplantsthatweremutantforeachofthetenAGOgenes.Theviralsymptomsonmostofthe plantswerethesameasonwildtypeplantsalthoughtheago2mutantsweremarkedlyhyper-susceptibletothisvirus.ago2 plantswerealsohyper-susceptibletocucumbermosaicvirus(CMV),confirmingthattheantiviralroleofAGO2isnotspecific toasinglevirus.Forbothviruses,thisphenotypewasassociatedwithtransientincreaseinvirusaccumulation.Inwildtype plantstheAGO2proteinwasinducedbyTCVandCMVinfection. Conclusions/Significance:BasedontheseresultsweproposethattherearemultiplelayerstoRNA-mediateddefenseand counter-defense in the interactions between plants and their viruses. AGO1 represents a first layer. With some viruses, includingTCVandCMV,thislayerisovercomebyviralsuppressorsofsilencingthatcantargetAGO1andasecondlayer involvingAGO2limitsvirusaccumulation.ThesecondlayerisactivatedwhenthefirstlayerissuppressedbecauseAGO2is repressedbyAGO1viamiR403.Theactivationofthesecondlayeristhereforeadirectconsequenceofthelossofthefirst layerofdefense.
