We demonstratedthathsa-miR-96issignificantlyandconsistentlyup-regulatedinall6NSCLCs.Wevalidatedthisresultinan independentsetof35pairedtumorsandtheiradjacentnormaltissues,aswellastheirserathatarecollectedbeforesurgical resectionorchemotherapy,andtheresultssuggestedthathsa-miR-96mayplayanimportantroleinNSCLCdevelopmentand hasgreatpotentialtobeusedasanoninvasivemarkerfordiagnosingNSCLC.WepredictedpotentialmiRNAtargetmRNAs based on different methods (TargetScan and miRanda). Further classification of miRNA regulated genes based on their relationshipwithmiRNAsrevealedthathsa-miR-96andcertainothermiRNAstendtodown-regulatetheirtargetmRNAsin NSCLCdevelopment,whichhaveexpressionlevelspermissivetodirectinteractionbetweenmiRNAsandtheirtargetmRNAs. Inaddition,weidentifiedasignificantcorrelationofmiRNAregulationwithgenescoincidewithhighdensityofCpGislands, which suggests that miRNA may represent a primary regulatory mechanism governing basic cellular functions and cell differentiations,andsuchmechanismmaybecomplementarytoDNAmethylationinrepressingoractivatinggeneexpression.
