AnalysisoftheBarA/UvrYTwo-ComponentSystemin ShewanellaoneidensisMR-1 LucasBinnenkade.,Ju¨rgenLassak.¤,KaiM.Thormann* DepartmentofEcophysiology,Max-Planck-Institutfu¨rTerrestrischeMikrobiologie,Marburg,Germany Abstract The BarA/UvrY two-component system is well conserved in species of the c-proteobacteria and regulates numerous processespredominantlybycontrollingtheexpressionofasubsetofnoncodingsmallRNAs.Inthisstudy,weidentifiedand characterizedtheBarA/UvrYtwo-componentsysteminthegammaproteobacteriumShewanellaoneidensisMR-1.Functional interaction of sensor kinase BarA and the cognate response regulator UvrY was indicated by in vitro phosphotransfer studies. The expression of two predicted small regulatory RNAs (sRNAs), CsrB1 and CsrB2, was dependent on UvrY. TranscriptomicanalysisbymicroarraysrevealedthatUvrYisaglobalregulatoranddirectlyorindirectlyaffectstranscript levelsofmorethan200genesinS.oneidensis.Amongthesearegenesencodingkeyenzymesofcentralcarbonmetabolism suchasackA,aceAB,andpflAB.Aspredictedofasignaltransductionpathwaythatcontrolsaspectsofcentralmetabolism, mutantslacking UvrY reach asignificantly higher ODthan thewild type during aerobic growthon N-acetylglucosamine (NAG)whileunderanaerobicconditionsthemutantgrewmoreslowly.Ashorterlagphaseoccurredwithlactateascarbon source.Incontrast,significantgrowthphenotypeswereabsentincomplexmedium.Basedonthesestudieswehypothesize that,inS.oneidensisMR-1,theglobalBarA/UvrY/Csrregulatorypathwayisinvolvedincentralcarbonmetabolismprocesses.
