Based on its function in vascular integrity, we hypothesize that the angiopoietin (Ang)-Tie2 system plays a role in the development of VILI. The present study was designed to examine the effects of mechanical ventilationontheAng-Tie2systeminlungtissue.Moreover,weevaluatedwhethertreatmentwithAng-1,aTie2receptor agonist,protectsagainstinflammation,vascularleakageandimpairedgasexchangeinducedbymechanicalventilation. Methods: Mice were anesthetized, tracheotomized and mechanically ventilated for 5hours with either an inspiratory pressureof10cmH O(‘low’tidalvolume,7.5ml/kg;LV )or18cmH O(‘high’tidalvolume,15ml/kg;HV ).Atinitiation 2 T 2 T ofHVT-ventilation,recombinanthumanAng-1wasintravenouslyadministered(1or4 mgperanimal).Non-ventilatedmice servedascontrols.
