We tested prion gene (PRNP) haplotypes for an associationwithatypicalBSE. Methodology/PrincipleFindings:HaplotypetaggingpolymorphismsthatcharacterizePRNPhaplotypesfromthepromoter region through thethreeprimeuntranslated region ofexon3 (25.2kb)wereused todetermine PRNPhaplotypesof six availableatypicalBSEcasesfromCanada,FranceandtheUnitedStates.Oneortwocopiesofadistinct PRNPhaplotype wereidentifiedinfiveofthesixcases(p=1.361024,two-tailedFisher’sexacttest;CI95% 0.263–0.901,differencebetween proportions).ThehaplotypespansaportionofPRNPthatincludespartofintron2,theentirecodingregionofexon3and partofthethreeprimeuntranslatedregionofexon3(13kb). Conclusions/Significance:ThisresultsuggeststhatageneticdeterminantinornearPRNPmayinfluencesusceptibilityof cattletoatypicalBSE.
