Genetically encoded sensors for cGMP have been developed based on fluorescence resonance 2+ energytransfer(FRET)betweenfluorescentproteins.However,toanalyzethedynamicrelationshipamongthesesecond messengers,combineduseofexistingsensorsinasinglecellisinadequatebecauseofthesignificantspectraloverlaps.A single wavelength indicator is an effective alternative to avoid this problem, but color variants of a single fluorescent protein-basedbiosensorarelimited.Inthisstudy,toconstructanewcolorfluorescentsensor,weconvertedtheFRET-based sensorintoasinglewavelengthindicatorusingadarkFRETacceptor.WedevelopedabluefluorescentcGMPbiosensor, whichisspectrallycompatiblewithaFRET-basedcAMPsensorusingcyanandyellowfluorescentproteins(CFP/YFP).We 2+ cotransfectedthemandloadedaredfluorescentprobeforCa intocells,andaccomplishedtriple-parameterfluorescence imagingofthesecyclicnucleotidesandCa ,confirmingtheapplicabilityofthiscombinationtoindividuallymonitortheir 2+ dynamics in a single cell.
