DifferentialDependenceonBeclin1fortheRegulation ofPro-SurvivalAutophagybyBcl-2andBcl-xLinHCT116 ColorectalCancerCells MurielPriault1,2*,ErikaHue3,4,FannyMarhuenda3,4,PaulPilet5,LisaOliver3,4,Franc?oisM.Vallette3,4 1CNRS IBGC UMR 5095, Bordeaux, France, 2Universite′ Victor Se′galen Bordeaux 2, Bordeaux, France, 3INSERM UMR 892, Nantes, France, 4Faculte′ de Me′decine, Universite′ deNantes,Nantes,France,5INSERMUMR791,Nantes,France Abstract Autophagyisdescribedtobeinvolvedinhomeostasis,developmentanddisease,bothasasurvivalandadeathprocess.Its involvementincelldeathproceedsfrominterrelationshipswiththeapoptoticpathway.Wefocusedonsurvivalautophagy andinvestigateditsinterplayswiththeapoptoticmachinery.WefoundthatwhileMcl-1remainedineffective,Bcl-2andBcl- xL were required for starved cells to display a fully functional autophagic pathway as shown by proteolysis activity and detectionofautophagicvesicles.Suchpro-autophagicfunctionsofBcl-2andBcl-xLwereindependentofBax.Howeverthey appearedtooperatethroughnonredundantmechanismsasBcl-xLwieldedatightercontrolthanBcl-2overtheregulation of autophagy: unlike Bcl-2, Bcl-xL and Atg7 manipulation yielded identical phenotypes suggesting they could be componentsofthesamesignallingpathway;Bcl-xLsubcellularlocalisationwasmodifieduponstarvation,andimportantly Bcl-xLactedindependentlyofBeclin1.StillanintactBH3-bindingsitewasrequiredforBcl-xLtostimulateafullyfunctional autophagicpathway.Thisstudyhighlightsthat,inadditiontotheirwell-establishedanti-deathfunctionduringapoptosis, Bcl-2andBcl-xLhaveabroaderroleincellsurvival.ShouldBcl-2andBcl-xLstandatthecross-roadsbetweenpro-survival andpro-deathautophagy,thisstudyintroducesthenewconceptthattheregulationofautophagybyBcl-2andBcl-xLis adjustedaccordingtoitssurvivalordeathoutcome.
