DisinhibitionMediatesaFormofHippocampalLong- TermPotentiationinAreaCA1 JakeOrmond*,MelanieA.Woodin* DepartmentofCellSystemsBiology,UniversityofToronto,Toronto,Ontario,Canada Abstract Thehippocampusplaysacentralroleinmemoryformationinthemammalianbrain.Itsabilitytoencodeinformationisthought to depend on the plasticity of synaptic connections between neurons. In the pyramidal neurons constituting the primary hippocampaloutputtothecortex,locatedinareaCA1,firingofpresynapticCA3pyramidalneuronsproducesmonosynaptic excitatorypostsynapticpotentials(EPSPs)followedrapidlybyfeedforward(disynaptic)inhibitorypostsynapticpotentials(IPSPs). Long-termpotentiation(LTP)ofthemonosynapticglutamatergicinputshasbecometheleadingmodelofsynapticplasticity,in partduetoitsdependenceonNMDAreceptors(NMDARs),requiredforspatialandtemporallearninginintactanimals.Using whole-cellrecordinginhippocampalslicesfromadultrats,wefindthattheefficacyofsynaptictransmissionfromCA3toCA1can beenhancedwithouttheinductionofclassicLTPattheglutamatergicinputs.Takingcarenottodirectlystimulateinhibitory fibers,weshowthattheinductionofGABAergicplasticityatfeedforwardinhibitoryinputsresultsinthereducedshuntingof excitatorycurrents,producingalong-termincreaseintheamplitudeofSchaffercollateral-mediatedpostsynapticpotentials.Like classicLTP,disinhibition-mediatedLTPrequiresNMDARactivation,suggestingaroleintypesoflearningandmemoryattributed primarilytotheformerandraisingthepossibilityofapreviouslyunrecognizedtargetfortherapeuticinterventionindisorders linkedtomemorydeficits,aswellasapotentiallyoverlookedsiteofLTPexpressioninotherareasofthebrain.
