Abstract
Adaptorprotein(AP)complexesregulateclathrin-coatedvesicleassembly,proteincargosorting,andvesiculartrafficking
betweenorganellesineukaryoticcells.BecausedisruptionofthevarioussubunitsoftheAPcomplexesisembryoniclethal
inthemajorityofcases,characterizationoftheirfunctioninvivoisstilllacking.Here,wedescribethefirstmutationinthe
humanAP1S1gene,encodingthesmallsubunits1AoftheAP-1complex.Thisfoundersplicemutation,whichleadstoa
premature stop codon, was found in four families with a unique syndrome characterized by mental retardation,
enteropathy,deafness,peripheralneuropathy,ichthyosis,andkeratodermia(MEDNIK).Tovalidatethepathogeniceffectof
themutation,weknockeddownAp1s1expressioninzebrafishusingselectiveantisensmorpholinooligonucleotides(AMO).
Theknockdownphenotypeconsistedofperturbationinskinformation,reducedpigmentation,andseveremotilitydeficits
duetoimpairedneuralnetworkdevelopment.Bothneuralandskindefectswererescuedbyco-injectionofAMOwithwild-
type(WT)humanAP1S1mRNA,butnotby
