DocosahexaenoicAcidforReading,Cognitionand BehaviorinChildrenAged7–9Years:ARandomized, ControlledTrial(TheDOLABStudy) AlexandraJ.Richardson*,JenniferR.Burton,RichardP.Sewell,TheesF.Spreckelsen,PaulMontgomery CentreforEvidence-BasedIntervention,UniversityofOxford,Oxford,UnitedKingdom Abstract Background:Omega-3fattyacidsaredietaryessentials,andthecurrentlowintakesinmostmoderndevelopedcountries arebelievedtocontributetoawidevarietyofphysicalandmentalhealthproblems.Evidencefromclinicaltrialsindicates thatdietarysupplementationwithlong-chainomega-3mayimprovechildbehaviorandlearning,althoughmostprevious trialshaveinvolvedchildrenwithneurodevelopmentaldisorderssuchasattention-deficit/hyperactivitydisorder(ADHD)or developmentalcoordinationdisorder(DCD).Hereweinvestigatedwhethersuchbenefitsmightextendtothegeneralchild population. Objectives:Todeterminetheeffectsofdietarysupplementationwiththelong-chainomega-3docosahexaenoicacid(DHA) onthereading,workingmemory,andbehaviorofhealthyschoolchildren. Design:Parallelgroup,fixed-dose,randomized,double-blind,placebo-controlledtrial(RCT). Setting:MainstreamprimaryschoolsinOxfordshire,UK(n=74). Participants: Healthychildrenaged 7–9years initiallyunderperforming in reading(#33rd centile). 1376 invited,362met studycriteria. Intervention:600mg/dayDHA(fromalgaloil),ortaste/colormatchedcorn/soybeanoilplacebo. Main Outcome Measures: Age-standardized measures of reading, working memory, and parent- and teacher-rated behavior. Results: ITT analyses showed no effect of DHA on reading in the full sample, but significant effects in the pre-planned subgroupof224childrenwhoseinitialreadingperformancewas#20thcentile(thetargetpopulationinouroriginalstudy design).Parent-ratedbehaviorproblems(ADHD-typesymptoms)weresignificantlyreducedbyactivetreatment,butlittleor noeffectswereseenforeitherteacher-ratedbehaviourorworkingmemory.
