DownregulationofChloroplastRPS1Negatively ModulatesNuclearHeat-ResponsiveExpressionofHsfA2 andItsTargetGenesinArabidopsis Hai-DongYu,Xiao-FeiYang,Si-TingChen,Yu-TingWang,Ji-KaiLi,QiShen,Xun-LiangLiu,Fang- QingGuo* TheNationalKeyLaboratoryofPlantMolecularGeneticsandNationalCenterforPlantGeneResearch(Shanghai),InstituteofPlantPhysiologyandEcology,Shanghai InstitutesforBiologicalSciences,ChineseAcademyofSciences,Shanghai,China Abstract Heatstresscommonlyleadstoinhibitionofphotosynthesisinhigherplants.Thetranscriptionalinductionofheatstress- responsivegenesrepresentsthefirstlineofinducibledefenseagainstimbalancesincellularhomeostasis.Althoughheat stresstranscriptionfactorHsfA2anditsdownstreamtargetgenesarewellstudied,theregulatorymechanismsbywhich HsfA2isactivatedinresponsetoheatstressremainelusive.Here,weshowthatchloroplastribosomalproteinS1(RPS1)isa heat-responsive protein and functions in protein biosynthesis in chloroplast. Knockdown of RPS1 expression in the rps1 mutantnearlyeliminatestheheatstress-activatedexpressionofHsfA2anditstargetgenes,leadingtoaconsiderablelossof heattolerance.WefurtherconfirmtherelationshipexistedbetweenthedownregulationofRPS1expressionandthelossof heat tolerance by generating RNA interference-transgenic lines of RPS1. Consistent with the notion that the inhibited activationofHsfA2inresponse toheatstressintherps1 mutantcausesheat-susceptibility, wefurther demonstratethat overexpressionofHsfA2withaviralpromoterleadstoconstitutiveexpressionsofitstargetgenesintherps1mutant,which is sufficient to reestablish lost heat tolerance and recovers heat-susceptible thylakoid stability to wild-type levels. Our findingsrevealaheat-responsiveretrogradepathwayinwhichchloroplasttranslationcapacityisacriticalfactorinheat- responsive activation of HsfA2 and its target genes required for cellular homeostasis under heat stress. Thus, RPS1 is an essentialyetpreviouslyunknowndeterminantinvolvedinretrogradeactivationofheatstressresponsesinhigherplants. Cit
