Method: Sixty-eight patients with nT2DM (nT2DM group), and 52 gender-, age- and body mass index (BMI) -matched normalglucosetolerance(NGTgroup)controlsparticipatedinthestudy.30nT2DMpatientswithFBG$14.0mmol/Lwere treatedwithCSIIfor2weeks,andwereunderwentaeuglycemic–hyperinsulinemicclamppre-andpost-treatment.Plasma FGF-21 concentrations were measured with a commercial ELISA kit. The relationship between plasma FGF-21 levels and metabolicparameterswasalsoanalyzed. Results:FastingplasmaFGF-21levelswerehigherinthenT2DMgroupthaninNGTgroups(1.6060.08vs.1.1360.26mg/L, P,0.01).InnT2DMpatients,fastingplasmaFGF-21concentrationsweresignificantlydecreasedafterCSIItreatmentfor2 weeks(1.6060.08vs.1.3060.05mg/L,P,0.05),accompaniedbyasignificantincreaseinthewholebodyglucoseuptake(M value) and blood glucose control. The changes in plasma FGF-21 levels (DFGF-21) were positively associated with the ameliorationofinsulinresistanceshownbythechangesinMvalue. Conclusion:PlasmaFGF-21levelisassociatedwithwholebodyinsulinsensitivityandsignificantlyreducedfollowingshort- termCSIItreatment.
