FlexibilityofaEukaryoticLipidome–InsightsfromYeast Lipidomics ChristianKlose.,MichalA.Surma.,MathiasJ.Gerl¤,FelixMeyenhofer,AndrejShevchenko,KaiSimons* MaxPlanckInstituteofMolecularCellBiologyandGenetics,Dresden,Germany Abstract Massspectrometry-basedshotgunlipidomicshasenabledthequantitativeandcomprehensiveassessmentofcellularlipid compositions.TheyeastSaccharomycescerevisiaehasproventobeaparticularlyvaluableexperimentalsystemforstudying lipid-relatedcellularprocesses.Here,byapplyingourshotgunlipidomicsplatform,weinvestigatedtheinfluenceofavariety of commonly used growth conditions on the yeast lipidome, including glycerophospholipids, triglycerides, ergosterol as wellascomplexsphingolipids.Thisextensivedatasetallowedforaquantitativedescriptionoftheintrinsicflexibilityofa eukaryoticlipidome,therebyprovidingnewinsightsintotheadjustmentsoflipidbiosyntheticpathways.Inaddition,we establishedabaselineforfuturelipidomicexperimentsinyeast.Finally,flexibilityoflipidomicfeaturesisproposedasanew parameterforthedescriptionofthephysiologicalstateofanorganism.
