GeneDistiller—DistillingCandidateGenesfromLinkage Intervals DominikSeelow1,JanaMarieSchwarz1,MarkusSchuelke1,2* 1DepartmentofNeuropaediatrics,Charite′ UniversityMedicalSchool,Berlin,Germany,2NeuroCureClinicalResearchCenter,Charite′ UniversityMedicalSchool,Berlin, Germany Abstract Background:Linkagestudiesoftenyieldintervalscontainingseveralhundredpositionalcandidategenes.Differentmanual orautomaticapproachesexistforthedeterminationofthegenemostlikelytocausethedisease.Whilethemanualsearchis veryflexibleandtakesadvantageoftheresearchers’backgroundknowledgeandintuition,itmaybeverycumbersometo collectandstudytherelevantdata.Automaticsolutionsontheotherhandusuallyfocusoncertainmodels,remain‘‘black boxes’’anddonotofferthesamedegreeofflexibility. Methodology: We have developed a web-based application that combines the advantages of both approaches. Informationfromvariousdatasourcessuchasgene-phenotypeassociations,geneexpressionpatternsandprotein-protein interactions was integrated into a central database. Researchers can select which information for the genes within a candidate interval or for single genes shall be displayed. Genes can also interactively be filtered, sorted and prioritised accordingtocriteriaderivedfromthebackgroundknowledgeandpreconceptionofthediseaseunderscrutiny. Conclusions: GeneDistiller provides knowledge-driven, fully interactive and intuitive access to multiple data sources. It displaysmaximumrelevantinformation,whilesavingtheuserfromdrowninginthefloodofdata.Atypicalquerytakesless thantwoseconds,thusallowinganinteractiveandexplorativeapproachtothehuntforthecandidategene.
