However, there is other evidenceshowingthatProm1-negativecancercellsalsoformtumorsinvivo.ThusitremainscontroversialwhetherProm1is abonafidemarkerforCSCs.ToverifyifProm1-expressingcellsareessentialfortumorigenesis,weestablishedamouseline, whoseProm1-expressingcellscanbeeliminatedconditionallybyaCre-inducibleDTAgeneontheProm1locustogether withatamoxifen-inducibleCreERTM,andgeneratedglioma-initiatingcells(GICs-LD)byoverexpressingboththeSV40Large TantigenandanoncogenicH-RasL61inneuralstemcellsofthemouseline.Weshowherethatthetamoxifen-treatedGICs- LD(GICs-DTA)formtumor-spheresincultureandtransplantableGBMinvivo.Thus,ourstudiesdemonstratethatProm1- expressingcellsaredispensableforgliomagenesisinthismousemodel.
