In this work,weperformedastudyaimedatanalyzingtheoriginsanddiversificationpatternsofanHCVoutbreakinWheelwright, asmallruraltownlocatedinSantaFeprovince(Argentina). Atotalof89outof1814bloodsamplescollectedfrompeople livinginWheelwright,werepositiveforHCVinfection.Thehighestprevalence(4.9%)wasobservedinpeopleolderthan50 years,withthehighestlevelforthegroupagedbetween70–79years(22%).TheRFLPanalysesshowedthat91%ofthe positivesamplesbelongedtotheHCV-1bgenotype.TheE1/E2andNS5Bgenesweresequenced,andtheirphylogenetic analysis showed that the HCV-1b sequences from Wheelwright were monophyletic. Bayesian coalescent-based methods were used to estimate substitution rates and time of the most recent common ancestor (tMRCA). The mean estimated substitutionratesandthetMRCAforE1/E2withandwithoutHVR1andNS5Bwere7.41E-03s/s/yand61years,5.05E-03s/s/ yand58yearsand3.24E-03s/s/yand53years,respectively.Insummary,thetMRCAvalues,thedemographicmodelwith constantpopulationsize,andthefactthatthehighestprevalenceofinfectionwasobservedinelderpeoplesupportthe hypothesis that the HCV-1b introduction in Wheelwright initially occurred at least five decade
