Ltd,Jinan,Shandong,China Abstract Background:ThehighlypathogenicavianinfluenzaH5N1viruscaninfectavarietyofanimalsandcontinuallyposesathreat toanimalandhumanhealth.WhilemanygenotypesofH5N1viruscanbefoundinchicken,fewareassociatedwiththe infectionofmammals.Characterizationofthegenotypesofviralstrainsinanimalpopulationsisimportanttounderstand the distribution of different viral strains in various hosts. This also facilitates the surveillance and detection of possible emergenceofhighlypathogenicstrainsofspecificgenotypesfromunknownhostsorhoststhathavenotbeenpreviously reportedtocarrythesegenotypes. Methodology/PrincipalFindings:TwoH5N1isolateswereobtainedfromlungsamplesoftworaccoondogsthathaddied fromrespiratorydiseaseinChina.Pathogenicityexperimentsshowedthattheisolateswerehighlypathogenictochicken. To characterize the genotypes of these viruses, their genomic sequences were determined and analyzed. The genetic contentsoftheseisolatesarevirtuallyidenticalandtheymaycomefromthesameprogenitorvirus.Phylogeneticanalysis indicatedthattheisolatesweregeneticallycloselyrelatedtogenotypeVH5N1virus,whichwasfirstisolatedinChinain 2003,andweredistinctfromthedominantvirusgenotypes(e.g.genotypeZ)ofrecentyears.Theisolatesalsocontaina multibasicaminoacidmotifattheirHAcleavagesitesandhaveanEresidueatposition627ofthePB2pr
