Pax7LineageContributionstotheMammalianNeural Crest BarbaraMurdoch1,2,CaseyDelConte2,Mart?′nI.Garc?′a-Castro2* 1BiologyDepartment,EasternConnecticutStateUniversity,Willimantic,Connecticut,UnitedStatesofAmerica,2DepartmentofMolecular,CellularandDevelopmental Biology,YaleUniversity,NewHaven,Connecticut,UnitedStatesofAmerica Abstract Background:Neuralcrestcellsarevertebrate-specificmultipotentcellsthatcontributetoavarietyoftissuesincludingthe peripheralnervoussystem,melanocytes,andcraniofacialbonesandcartilage.Abnormaldevelopmentoftheneuralcrestis associated with several human maladies including cleft/lip palate, aggressive cancers such as melanoma and neuroblastoma,andraresyndromes,likeWaardenburgsyndrome,acomplexdisorderinvolvinghearinglossandpigment defects.WepreviouslyidentifiedthetranscriptionfactorPax7asanearlymarker,andrequiredcomponentforneuralcrest developmentinchickembryos.Inmammals,Pax7isalsothoughttoplayaroleinneuralcrestdevelopment,yettheprecise contributionofPax7progenitorstotheneuralcrestlineagehasnotbeendetermined. Methodology/PrincipalFindings:HereweuseCre/loxPtechnologyindoubletransgenicmicetofatemapthePax7lineage inneural crestderivates.Wefind thatPax7descendantscontribute tomultipletissuesincludingthecranial,cardiacand trunkneuralcrest,whichinthecranialcartilageformadistinctregionalpattern.ThePax7lineage,likethePax3lineage,is additionallydetectedinsomenon-neuralcresttissues,includingasubsetoftheepithelialcellsinspecificorgans. Conclusions/Significance: These results demonstrate a previously unappreciated widespread distribution of Pax7 descendants withinandbeyondtheneural crest. Theyshedlight regardingtheregionallydistinctphenotypes observed in Pax3 and Pax7 mutants, and provide a unique perspective into the potential roles of Pax7 during disease and development.
