JiangandLiuBMCPlantBiology2010,10:40 /1471-2229/10/40 RESEARCH ARTICLE OpenAccess Pb-inducedcellulardefensesystemintheroot meristematiccellsofAlliumsativumL WushengJiang1,DonghuaLiu2* Abstract Background:Electronmicroscopy(EM)techniquesenableidentificationofthemainaccumulationsoflead(Pb)in cellsandcellularorganellesandobservationsofchangesincellultrastructure.Althoughthereisextensiveliterature relatingtostudiesontheinfluenceofheavymetalsonplants,Pbtolerancestrategiesofplantshavenotyetbeen fullyexplained.AlliumsativumL.isapotentialplantforabsorptionandaccumulationofheavymetals.Inprevious investigationstheeffectsofdifferentconcentrations(10-5 to10-3 M)ofPbwereinvestigatedinA.sativum, indicatingasignificantinhibitoryeffectonshootandrootgrowthat10-3 to10-4 MPb.Inthepresentstudy,we usedEMandcytochemistrytoinvestigateultrastructuralalterations,identifythesynthesisanddistributionof cysteine-richproteinsinducedbyPbandexplainthepossiblemechanismsofthePb-inducedcellulardefense systeminA.sativum. Results:After1hofPbtreatment,dictyosomeswereaccompaniedbynumerousvesicleswithincytoplasm.The endoplasmreticulum(ER)withswollencisternaewasarrangedalongthecellwallafter2h.Someflattened cisternaewerebrokenupintosmallclosedvesiclesandthenuclearenvelopewasgenerallymoredilatedafter4h. During24-36h,phenomenaappearedsuchashighvacuolizationofcytoplasmandelectron-densegranulesincell walls,vacuoles,cytoplasmandmitochondrialmembranes.Otherchangesincludedmitochondrialswellingandloss ofcristae,andvacuolizationofERanddictyosomesduring48-72h.InthePb-treatmentgroups,silvergrainswere observedincellwallsandincytoplasm,suggestingtheGomori-SwiftreactioncanindirectlyevaluatethePbeffects onplantcells. Conclusions:CellwallscanimmobilizesomePbions.Cysteine-richproteinsincellwallswereconfirmedbythe Gomori-Swiftreaction.Themorphologicalalterationsinplasmamembrane,dictyosomesandERreflectthefeatures ofdetoxificationandtoleranceunderPbstress.VacuolesareultimatelyoneofmainstoragesitesofPb.Root meristem
