PeripheralBloodInvariantNaturalKillerTCellsofPig- TailedMacaques XiangmingLi1,PatriciaPolacino2,RaquelGarcia-Navarro1,Shiu-LokHu2,3,MoriyaTsuji1* 1HIV and Malaria Vaccine Program, Aaron Diamond AIDS Research Center, Affiliate of the Rockefeller University, New York, New York, United States of America, 2WashingtonNationalPrimateResearchCenter,UniversityofWashington,Seattle,Washington,UnitedStatesofAmerica,3DepartmentofPharmaceutics,Universityof Washington,Seattle,Washington,UnitedStatesofAmerica Abstract In humans, invariant natural killer T (iNKT) cells represent a small but significant population of peripheral blood mononuclearcells(PBMCs)withahighdegreeofvariability.Inthisstudy,pursuanttoourgoalofidentifyinganappropriate non-humanprimatemodelsuitableforpre-clinical glycolipidtesting,weevaluatedthepercentageandfunctionof iNKT cellsintheperipheralbloodofpig-tailedmacaques.First,usingahumanCD1d-tetramerloadedwitha-GalCer(a-GalCer- CD1d-Tet),wefoundthata-GalCer-CD1d-Tet+CD3 iNKTcellsmakeup0.13%to0.4%ofpig-tailedmacaquePBMCs,which arecomparabletothepercentageofiNKTcellsfoundinhumanPBMCs.Second,weobservedthatalargeproportionof + Va24 CD3 cellsarea-GalCer-CD1d-Tet+CD3 iNKTcells,whichprimarilyconsistofeithertheCD4 orCD8 subpopulation. Third,wefoundthatpig-tailedmacaqueiNKTcellsproduceIFN-cinresponsetoa-GalCer,asshownbyELISpotassayand intracellularcytokinestaining(ICCS),aswellasTNF-a,asshownbyICCS,indicatingthattheseiNKTcellsarefullyfunctional. + + + + + + Interestingly,themajorityofpig-tailedmacaqueiNKTcellsthatsecreteIFN-careCD8 iNKTcells.Basedonthesefindings, weconclude that thepig-tailed macaques exhibit potential as anon-humananimal modelfor thepre-clinical testing of iNKT-stimulatingglycolipids.
