DNA vaccination,anoveltherapeuticstrategyforchronicvirusinfection, hasbeenshowntoinduceCTLresponses.However, accumulateddatahaveshownthatCTLscouldnotbeeffectivelyinducedbyHBVDNAvaccination. Methodology/PrincipalFindings:Here,wereportthatpraziquantel(PZQ),ananti-schistomadrug,couldactasanadjuvant to overcome the lack of potent CTL responses by HBV DNA vaccination in mice. PZQ in combination with HBV DNA + vaccinationaugmentedtheinductionofCD8 Tcell-dependentandHBV-specificdelayedhypersensitivityresponses(DTH) inC57BL/6mice.Furthermore,theinducedCD8 TcellsconsistedofbothTc1andTc17subtypes.ByusingIFN-cknockout + (KO)miceandIL-17KOmice,bothcytokineswerefoundtobeinvolvedintheDTH.TherelevanceofthesefindingstoHBV immunizationwasestablishedinHBsAgtransgenicmice,inwhichPZQalsoaugment
