Methods:Atotalof501E.coliisolatesfrompigfeces,pigcarcassesandhumanstoolsweretestedfortheir susceptibilitytoselectedantimicrobial.MultiplexPCRwasconductedtodetectthepresenceofthreesulgenes amongthesulfonamide-resistantE.coliisolates.Fifty-sevensulfonamide-resistantE.coliwereselectedbasedon presenceofsulresistancegenesandsubjectedtoconjugationand/ortransformationexperiments.S1nuclease digestionfollowedbypulsed-fieldgelelectrophoresiswasusedtovisualizeanddeterminethesizeofplasmids. PlasmidscarryingsulgeneswerecharacterizedbyPCR-basedreplicontypingtoallowacomparisonofthetypesof sulgenes,thereservoirandplasmidpresent. Results:Atotalof109/501isolatesexhibitedsulfonamideresistance.Therelativeprevalencesofsulgenesfromthe threereservoirs(pigs,pigcarcassesandhumans)were65%,45%and12%forsul2,sul1,andsul3 ,respectively. Transferofresistancethroughconjugationwasobservedin42/57isolates.Resistancestostreptomycin,ampicillin andtrimethoprimwereco-transferredinmoststrains.Class1integronswerepresentin80%ofsul1-carrying plasmidsand100%ofsul3-carryingplasmids,butonlyin5%ofsul2-carryingplasmids.Thesulplasmidsranged from33to160-kbinsizeandbelongedtoninedifferentincompatibility(Inc)groups:FII,FIB,I1,FIA,B/O,FIC,N, HI1andX1.IncFIIwasthedominanttypeinsul2-carryingplasmids(52%),whileIncI1wasthemostcommontype insul1andsul3-carryingplasmids(33%and45%,respectively).Multirepliconswerefoundassociatedwithallthree sulgenes.
