Methods:TheeffectsofPienTzeHuangonbrainproteinexpressioninspontaneouslyhypertensiverat(SHR)and strokeproneSHR(SHRsp)werestudiedwith2-Dgelelectrophoresisandmassspectrometricanalysiswithamatrix- assistedlaserdesorption/ionizationtime-of-flight(MALDI-TOF)/TOFtandemmassspectrometerandonbraincell deathwithenzymelinkimmunosorbentassay(ELISA)andimmunostaining. Results:PienTzeHuangdecreasedcelldeathinhippocampusandcerebellumcausedbychronicischemiaand hypertensivestroke.Immunostainingofcaspase-3resultsindicatedthatPienTzeHuangpreventsbraincellsfrom apoptosiscausedbyischemia.BrainproteinexpressionresultssuggestedthatPienTzeHuangdownregulated QCR2intheelectrontransferchainofmitochondriapreventingreactiveoxygenspecies(ROS)damageandpossibly subsequentcelldeath(caspase3assay)ascausedbychronicischemiaorhypertensivestroketohippocampusand cerebellum. Conclusion:PienTzeHuangshowedpreventiveeffectsonlimitingthedamageorinjurycausedbychronic ischemiaandhypertensivestrokeinrats.TheeffectofPienTzeHuangwaspossiblyrelatedtopreventionofcell deathfromapoptosisorROS/oxidativedamageinmitochondria.
