Cells of budding yeast (Saccharomyces cerevisiae) lacking Rtt101p, a relative of the E3 ubiquitin-ligase components Cul3 and Cul4, display a delay in the metaphase-to-anaphase transition of mitosis and show increased sensitivity to genotoxic drugs. These problems can be traced back to a defect in replicating certain regions of the genome - ‘slow zones’ - which include the rRNA locus. In these areas, Rtt101p appears to be required for the restart of collapsed replication forks (but not of the ones that have only stalled but are still intact), suggesting a role for ubiquitination and possibly protein degradation in this process.
