PsychedelicsandtheHumanReceptorome ThomasS.Ray* DepartmentofZoology,UniversityofOklahoma,Norman,Oklahoma,UnitedStatesofAmerica Abstract We currently understand the mentaleffects ofpsychedelics to be causedby agonism orpartial agonism of 5-HT2A (and possibly5-HT2C)receptors,andweunderstandthatpsychedelicdrugs,especiallyphenylalkylamines,arefairlyselectivefor thesetworeceptors.Thismanuscriptisareferenceworkonthereceptoraffinitypharmacologyofpsychedelicdrugs.New data is presented on the affinity of twenty-five psychedelic drugs at fifty-one receptors, transporters, and ion channels, assayed by the National Institute of Mental Health – Psychoactive Drug Screening Program (NIMH-PDSP). In addition, comparabledatagatheredfromtheliteratureontenadditionaldrugsisalsopresented(mostlyassayedbytheNIMH-PDSP). Anewmethodisintroducedfornormalizingaffinity(Ki)datathatfactorsoutpotencysothatthemulti-receptoraffinity profilesofdifferentdrugscanbedirectlycomparedandcontrasted.Themethodisthenusedtocomparethethirty-five drugsingraphicalandtabularform.Itisshownthatpsychedelicdrugs,especiallyphenylalkylamines,arenotasselectiveas generallybelieved,interactingwithforty-twoofforty-ninebroadlyassayedsites.Thethirty-fivedrugsofthestudyhavevery diversepatternsofinteractionwithdifferentclassesofreceptors,emphasizingeighteendifferentreceptors.Thisdiversityof receptorinteractionmayunderliethequalitativediversityofthesedrugs.Itshouldbepossibletousethisdiversesetof drugsasprobesintotherolesplayedbythevariousreceptorsystemsinthehumanmind.
