To advance the knowledge of serological immune responses against and to assess the scope of tumor antigenicityofcutaneouslymphoma,1-and2-dimensionalWesternblotanalyseswithserafrompatientswerecombined withproteomics-basedproteinidentification.Testingserafrom87cutaneouslymphomapatientsby1-dimensionalWestern blotanalysis,64casesofseroreactivityagainstlymphomacellswerefound.Thepositiveresponseswererelativelyweak, restrictedtofewantigensineachcase,andheterogeneous.Toidentifytheantigens,proteinsofthemycosisfungoidescell lineMyLaandprimarytumorcellswereseparatedby2-dimensionalgelelectrophoresis,Western-blottedandprobedwith heterogeneous and autologous patient sera. The antigens were identified from silver-stained replica gels by MALDI-TOF massspectrometry.14differentantigenswereassignedandidentifiedwiththisproteome-serologicalapproach.Onlyone, vimentin,hadbeenreportedbefore,theother13arenewantigensforcutaneouslymphomas.
