QuantifyingAdaptiveEvolutionintheDrosophila ImmuneSystem DarrenJ.Obbard1*,JohnJ.Welch1,Kang-WookKim1,2,FrancisM.Jiggins3 1InstituteofEvolutionaryBiology,UniversityofEdinburgh,Edinburgh,UnitedKingdom,2DepartmentofAnimalandPlantSciences,UniversityofSheffield,Sheffield, UnitedKingdom,3DepartmentofGenetics,UniversityofCambridge,Cambridge,UnitedKingdom Abstract ItisestimatedthatalargeproportionofaminoacidsubstitutionsinDrosophilahavebeenfixedbynaturalselection,andas organisms are faced with an ever-changing array of pathogens and parasites to which they must adapt, we have investigatedtheroleofparasite-mediatedselectionasalikelycause.Toquantifytheeffect,andtoidentifywhichgenesand pathwaysaremostlikelytobeinvolvedinthehost–parasitearmsrace,wehavere-sequencedpopulationsamplesof136 immunity and 287 position-matched non-immunity genes in two species of Drosophila. Using these data, and a new extensionoftheMcDonald-Kreitmanapproach,weestimatethatnaturalselectionfixesadvantageousaminoacidchanges inimmunitygenesatnearlydoubletherateofothergenes.Wefindtherateofadaptiveevolutioninimmunitygenesisalso morevariablethanothergenes,withasmallsubsetofimmunegenesevolvingunderintenseselection.Thesegenes,which are likely to represent hotspots of host–parasite coevolution, tend to share similar functions or belong to the same pathways,suchastheantiviralRNAipathwayandtheIMDsignallingpathway.Thesepatternsappeartobegeneralfeatures ofimmunesystemevolutioninbothspecies,asratesofadaptiveevolutionarecorrelatedbetweentheD.melanogasterand D. simulans lineages. In summary, our data provide quantitative estimates of the elevated rate of adaptive evolution in immunesystemgenesrelativetotherestofthegenome,andtheysuggestthatadaptationtoparasitesisanimportant forcedrivingmolecularevolution.
