However, other protein classes might be involvedin[PSI ]formation. + Results: We designed a functional proteomic study that combines two techniques to identify modulators of Sup35p aggregationanddescribethechangesassociatedto[PSI ]formation.Thefirstallowsmeasuringtheeffectoffractionated + + Saccharomycescerevisiaecytosolicextractsfrom[PSI ]and[psi2]yeastcellsonSup35passembly.Thesecondisamultiplex qualitativeandquantitativecomparisonofproteincompositionofactiveandinactivefractionsusingagel-freeandlabel- free LC-MS approach. We identify changes in proteins involved in translation, folding, degradation, oxido-reduction and metabolicprocesses. Conclusion:Ourfunctionalproteomicstudyprovidesthefirstinventorylistofover300proteinsthatdirectlyorindirectly affectSup35paggregationand[PSI ]formation.Ourresultshighlightthecomplexityofthecellularchangesaccompanying + + [PSI ]formationandpavethewayforinvitrostudiesaimedtodocumenttheeffectofindividualand/orcombinationsof proteinsidentifiedhere,susceptibleofaffectingSup35passembly.
