Methodology/PrincipalFindings:WeinvestigatedQfeverinruralSenegal.Humansamples(e.g.,sera,saliva,breastmilk, feces)werescreenedinthegenerallyhealthypopulationoftwovillagesoftheSine-Saloumregion.Tickswerecollectedin fourregions.Seroprevalencewasstudiedbyimmunofluorescence,andallothersamplesweretestedbytwoqPCRsystems fordetectionofC.burnetii.Positivesamplesweregenotyped(multispacertyping)byamplificationandsequencingofthree spacers.Strainswereisolatedbycellculture.Wefoundthattheseroprevalencemaybeashighas24.5%(59of238studied) inDielmovillage.WeidentifiedspontaneousexcretionofC.burnetiibyhumansthroughfaecesandmilk.Hardandsoftticks (8species)wereinfectedin0–37.6%.WeidentifiedthreegenotypesofC.burnetii.Thepreviouslyidentifiedgenotype6was themostcommoninticksinallstudiedregionsandtheonlyonefoundinhumansamples.Threestrainsofgenotype6ofC. burnetii were also recovered from soft tick Ornithodoros sonrai. Two other genotypes found in ticks, 35 and 36, were identifiedforthefirsttime. Conclusions/Significance: QfevershouldbeconsideredasignificantpublichealththreatinSenegal.Humans,similarto othermammals,maycontinuouslyexcreteC.burnetii.
