Methods:HumansynovialfibroblastswerestimulatedwithMSUinthepresenceorabsenceofserumamyloidA (SAA).Thecellularsupernatantswereanalyzedbyimmunoblottingusinganti-IL-1boranti-caspase-1antibodies.IL- 1borNLRP3mRNAexpressionswereanalyzedbyreal-timePCRorreversetranscription-PCR(RT-PCR)method. Results:NeitherSAAnorMSUstimulationresultedinIL-1borinterleukin-1a(IL-1a)secretionsandpro-IL-1b processinginsynovialfibroblasts.However,inSAA-primedsynovialfibroblasts,MSUstimulationresultedinthe activationofcaspase-1andproductionofactiveIL-1bandIL-1a.TheeffectofSAAonIL-1binductionwas impairedincellsbysilencingNLRP3usingsiRNAortreatingwithcaspase-1inhibitor.Inaddition,SAAinducedthe secretionofcathepsinBandNLRP3mRNAexpressioninsynovialfibroblasts. Conclusions:OurdatademonstratethatexposureofhumansynovialfibroblaststoSAApromotesMSU-mediated caspase-1activationandIL-1bsecretionintheabsenceofmicrobialstimulation.Thesefindingsprovideinsightinto themolecularprocessesunderlyingthesynovialinflammatoryconditionofgout.
