Methods:Corticalastrocytecultureswerepreparedfrompostnatalday1(onedayafterbirth)maleorfemaleCD1 micepups.Inaddition,corticalastrocytecultureswerealsopreparedfromfemalepupsthatwereinjectedatbirth with100μgoftestosteronepropionateorvehicle.Culturesweretreatedfor5hourswithmediumcontaining lipopolysaccharide(LPS)orwithcontrolmedium.ThemRNAlevelsofIL6,interferon-inducibleprotein10(IP10), TNFa,IL1b,Toll-likereceptor4(TLR4),steroidogenicacuteregulatoryproteinandtranslocatorproteinwere assessedbyquantitativereal-timepolymerasechainreaction.Statisticalsignificancewasassessedbyunpairedt-test orbyone-wayanalysisofvariancefollowedbytheTukeyposthoctest. Results:ThemRNAlevelsofIL6,TNFaandIL1bafterLPStreatmentweresignificantlyhigherinastrocytesderived frommaleorandrogenizedfemalescomparedtoastrocytesderivedfromcontrolorvehicle-injectedfemales.In contrast,IP10mRNAlevelsafterLPStreatmentwerehigherinastrocytesderivedfromcontrolorvehicle-injected femalesthaninthoseobtainedfrommalesorandrogenizedfemales.Thedifferentresponseofmaleandfemale astrocytestoLPSwasdueneithertodifferencesinthebasalexpressionoftheinflammatorymoleculesnorto differencesintheexpressionoftheLPSreceptorTLR4.I
