Methods:Inordertoassesstheexvivoeffectsof17b-oestradiolinintacthumancardiactissues,wedevelopeda 24-hmodelforthecultureofhumanatrialmyocardium.Weverifiedtissueviabilityafter24hinculturewithtwo standardassaystodeterminethedegreeofapoptosisandmetabolicactivityofcardiactissues.Progesterone receptormRNAandproteinlevelweremeasuredafter24-htreatmentoftissueswith17b-oestradiol.Statistical analysiswasperformedbytheMann-WhitneyUtestandtwo-wayANOVA. Results:Weestablishedatissueculturemodelthatallowsforthestudyofviablehumancardiactissueovera24-h period.After24h,culturedcardiactissuesrevealedlowapoptosis,retainedtheirmetabolicactivityand,therefore, remainedviable.Treatmentwith17b-oestradiolledtoaninductionoftheprogesteronereceptormRNAlevelin female(P=0.001)butnotinmaletissues.Similarly,therewasanincreaseinthelevelofprogesteronereceptor proteininfemaletissues(P=0.03),whileadecreasingtrendwasobservedinmaletissues(P=0.079)exposedto 17b-oestradiol. Conclusions:Ournovelfindingmayofferamolecularexplanationforthesex-specificdifferencesobservedin cardiacremodelling.Theculturemodelweestablishedforhumancardiactissuewillfacilitatethestudyofcellular processesinhealthanddiseaseandwillbeofuseforpharmacologicaltesting.
