Methods:BALB/cmicereceivedasingleoracombinedintratrachealadministrationofC.albicans(1×105CFU/ mouse)andP.aeruginosa(1×107CFU/mouse)attime0(T0)uponC.albicanscolonization,andDay2.To evaluatetheeffectofantifungaltherapy,micereceivedcaspofunginintraperitoneallydaily,eitherfromT0orfrom Day1post-colonization.AftersacrificeatDay4,lungswereanalyzedforhistologicalscoring,measurementof endothelialinjury,andquantificationofliveP.aeruginosaandC.albicans.Bloodsampleswereculturedfor dissemination. Results:Asignificantdecreaseinlungendothelialpermeability,theamountofP.aeruginosa,andbronchiole inflammationwasobservedincaseofpriorC.albicanscolonization.Mortalityrateandbacterialdisseminationwere unchangedbypriorC.albicanscolonization.CaspofungintreatmentfromT0(notfromDay1)increasedtheirlevels ofendothelialpermeabilityandlungP.aeruginosaloadsimilarlytomicereceivingP.aeruginosaalone. Conclusions:P.aeruginosa-inducedlunginjuryisreducedwhenprecededbyshorttermC.albicansairway colonization.AntifungaldrugcaspofunginreversesthateffectwhenusedfromT0andnotfromDay1.
