Abstract
Background:Identificationofgenesthatmodulatelongevityisamajorfocusofaging-relatedresearchandanareaofintense
publicinterest.Inadditiontofacilitatinganimprovedunderstandingofthebasicmechanismsofaging,suchgenesrepresent
potentialtargetsfortherapeuticinterventioninmultipleage-associateddiseases,includingcancer,heartdisease,diabetes,and
neurodegenerativedisorders.Todate,however,targetedeffortsatidentifyinglongevity-associatedgeneshavebeenlimitedby
alackofpredictivepower,andusefulalgorithmsforcandidategene-identificationhavealsobeenlacking.
Methodology/PrincipalFindings:Wehaveutilizedashortest-pathnetworkanalysistoidentifynovelgenesthatmodulate
longevityinSaccharomycescerevisiae.Basedonasetofpreviouslyreportedgenesassociatedwithincreasedlifespan,we
applied a shortest-path network algorithm to a pre-existing protein–protein interaction dataset in order to construct a
shortest-pathlongevitynetwork.Tovalidatethisnetwork,thereplicativeagingpotentialof88single-genedeletionstrains
correspondingtopredictedcomponentsoftheshortest-pathlongevitynetworkwasdetermined.Herewereportthatthe
single-gene deletion strains identified by our shortest-path longevity analysis are significantly enriched for mutations
conferringeitherincreasedordecreasedreplicativelifespan,relativetoarandomlyselectedsetof564single-genedeletion
strainsortothecurrentdatasetavail
