Miles2,Bjo¨rnAndersson1 1ScienceforLifeLaboratory,DepartmentofCellandMolecularBiology,KarolinskaInstitutet,Stockholm,Sweden,2DepartmentofPathogenMolecularBiology,Faculty ofInfectiousandTropicalDiseases,LondonSchoolofHygieneandTropicalMedicine,London,UnitedKingdom Abstract TrypanosomacruziisthecausativeagentofChagasdisease,whichaffectsmorethan9millionpeopleinLatinAmerica.We havegenerated adraftgenomesequenceoftheTcIstrainSylvioX10/1 andcomparedittotheTcVIreferencestrain CL Brenerto identifylineage-specific features.We foundvirtually no differencesin thecoregene contentof CL Brenerand SylvioX10/1bypresence/absenceanalysis,but6openreadingframesfromCLBrenerweremissinginSylvioX10/1.Several multicopygenefamilies,includingDGF,mucin,MASPandGP63werefoundtocontainsubstantiallyfewergenesinSylvio X10/1,basedonsequencereadestimations.1,861smallinsertion-deletioneventsand77,349nucleotidedifferences,23%of which were non-synonymous and associated with radical amino acid changes, further distinguish these two genomes. Therewere336genesindicatedasunderpositiveselection,145uniquetoT.cruziincomparisontoT.bruceiandLeishmania. ThisstudyprovidesaframeworkforfurthercomparativeanalysesoftwomajorT.cruzilineagesandalsohighlightstheneed forsequencingmorestrainstounderstandfullythegenomiccompositionofthisparasite.
