PandeyandSoupirAMBExpress2011,1:18 /content/1/1/18 ORIGINAL OpenAccess Escherichiacoliinactivationkineticsinanaerobic digestionofdairymanureundermoderate, mesophilicandthermophilictemperatures PramodKPandey*andMichelleLSoupir Abstract Batchanaerobicdigestionexperimentsusingdairymanureasfeedstockswereperformedatmoderate(25°C), mesophilic(37°C),andthermophilic(52.5°C)temperaturestounderstandE.coli,anindicatororganismfor pathogens,inactivationindairymanure.Incubationperiodsat25,37,and52.5°C,were61,41,and28days respectively.ResultswereusedtodevelopmodelsforpredictingE.coliinactivationandsurvivalinanaerobic digestion.FormodelingweusedthedecayofE.coliateachtemperaturetocalculatethefirst-orderinactivation ratecoefficients,andtheserateswereusedtoformulatethetime-temperature-E.colisurvivalrelationships.We foundtheinactivationratecoefficientat52.5°Cwas17and15timeslargerthantheinactivationratecoefficientsat 25and37°C,respectively.Decimalreductiontimes(D10;timetoachieveonelogremoval)at25,37,and52.5°C, were9-10,7-8days,and1day,respectively.TheArrheniuscorrelationbetweeninactivationratecoefficients andtemperaturesovertherange25-52.5°CwasdevelopedtounderstandtheimpactsoftemperatureonE.coli inactivationrate.Usingthiscorrelation,thetime-temperature-E.colisurvivalrelationshipswerederived.Besides E.coliinactivation,impactsoftemperatureonbiogasproduction,methanecontent,pHchange,ORP,andsolid reductionwerealsostudied.Athighertemperatures,biogasproductionandmethanecontentwasgreaterthan thatatlowtemperatures.WhileatthermophilictemperaturepHwasincreased,atmesophilicandmoderate temperaturespHwerereducedovertheincubationperiod.Theseresultscanbeusedtounderstandpathogen inactivationduringanaerobicdigestionofdairymanure,andimpactsoftemperaturesonperformanceofanaerobic digesterstreatingdairymanure.
