2012;32(12) 南方医科大学学报（J South Med Univ ） ·1713 · 基础研究 柯萨奇病毒A 16型VP 1~VP4 基因克隆及其表达产物的抗原相关 性分析 1 2 2 1 1 2 1 宋远斌 ，何思杰 ，余 楠 ，陈欣欣 ，王 斌 ，车小燕 ，曾其毅 1 2 南方医科大学珠江医院 儿科中心，医学检验中心，广东 广州 510282 摘要：目的克隆并表达柯萨奇病毒A 16型VP 1~VP4 蛋白基因，初步分析其抗原相关性。方法抽提病毒RNA ，经RT-PCR 方法 分别扩增出VP 1~VP4 蛋白基因片段，经克隆后，在QIA 表达系统中表达，表达产物经8 mol/L 尿素洗涤及镍柱亲和层析纯化后， 用柯萨奇病毒A 16型病毒免疫兔血清及肠道病毒71 型病毒免疫兔血清对重组蛋白进行Western blotting 及ELISA 分析其抗原 相关性及交叉反应性。结果成功构建的重组质粒pQE30a/VP1~VP4 并经IPTG 诱导，重组蛋白VP 1~VP4 高效表达并纯化成 功，经Western blotting 及ELISA 证实重组蛋白VP 1~VP4 可以被CVA16免疫兔血清特异识别，部分与肠道病毒71 型免疫血清存 在交叉反应。结论在大肠杆菌M 15 中高效表达出柯萨奇病毒A 16型VP 1~VP4 蛋白，经纯化产物具有较强的抗原反应性。 关键词：柯萨奇病毒A 16型；VP 1蛋白；VP2 蛋白；VP3蛋白；VP4 蛋白；克隆；原核表达；抗原反应性 中图分类号：R373.23 文献标志码：A 文章编号：1673-4254（2012）12-1713-05 doi: 10.3969/j.issn. 1673-4254.2012.12.007 Cloning, expression and antigenic analysis of VP1-VP4 gene encoding the structural protein of Coxsackie virus A16 1 2 2 1 1 2 1 SONG Yuanbin , HE Sijie , YU Nan , CHEN Xinxin , WANG Bin , CHE Xiaoyan , ZENG Qiyi 1Pediatric Center, 2Laboratory Testing Center, Zhujiang Hospital, Southern Medical University, Guangzhou 510282, China Abstract: Objective To clone and express VP1-VP4 genes encoding the structural proteins of Coxsackie virus A16 and analyze the antigenicity of the expressed recombinant proteins.
