婴幼儿配方奶粉中核苷酸含量的测定方法研究 杨大进 　方从容 　马 　兰 　王竹天 ( 中国疾控中心营养与食品安全所 ,北京 　10002 1) 摘 　要 :为检测婴幼儿配方奶粉中胞嘧啶核苷酸 、尿嘧啶核苷酸 、腺嘌呤核苷酸 、鸟嘌呤核苷酸 、次 黄嘌呤核苷酸 5 种核苷酸的含量 ,建立了奶粉中 5 种核苷酸的紫外检测器液相色谱法 。对试样提 取 、固相萃取等前处理方式和仪器检测条件进行了研究 ,试样经 05 % 乙酸去除蛋白质 ,氨基固相 萃取柱富集和净化 ,Symmetry C 柱分离 ,254 nm 紫外检测器液相色谱分析方法定量检测 。胞嘧啶 18 核苷酸 、尿嘧啶核苷酸 、腺嘌呤核苷酸 、鸟嘌呤核苷酸和次黄嘌呤核苷酸的添加 回收率分别是 900 %～942 % 、864 %～932 % 、857 %～926 % 、900 %和 857 %～933 % ,相对标准差分别为 μ 66 % 、34 % 、82 % 、68 %和 82 % ,检出限分别为 004 、005 、005 、006 和 004 gmL 。3 年来对 30 件送检样品的检测显示 ,检测结果与标示量基本一致 。该方法适用于婴幼儿配方奶粉核苷酸含 量的分析 。 关键词 :乳制品;婴儿食品;核苷酸类 ;色谱法 ,高压液相 A method for determination of nucleotides in infant formula milk powder Yang Daj in , et al . We investigated the pretreatment means including sample extraction ,solid phase extraction and conditions of instrumentation and established the method in which 05 % acetic acid was used for elimination of protein ,quaternary amine solid phase extraction column for concentration and purification , symmetry C18 column for isolation and the nucleotides were determined quantitatively at UV 254 nm wavelength with HPLC. Recovery rates of cytidine ,uridine ,adenosine ,guanosine and inosine were in ranges of 900 %～ 942 % 、864 % ～932 % 、857 % ～926 % 、900 % and 857 % ～933 % respectively , the RSDs were 66 % 、34 % 、82 % 、68 % and 82 % respectively ,the
