生物技术通报 · 研究报告 · BIOTECHNOLOGY BULLETlN 2008年第 1期 Bassam和Sanguinetti银染方法在SRAP和TRAP 标记中的比较研究 任民 贾兴华 蒋彩虹 杨爱国 王日新 (中国农业科学院烟草研究所，青岛 266101) 摘 要：银染作为一种重要的DNA染色方法，对分子标记检测有重要影响。SRAP标记和TRAP标记是两种较为 新型的分子标记，近年来得到了广泛应用，尤其在缺少遗传图谱的物种中应用价值更大。以普通烟草种(Nicotiana tabacum L．)烤烟和香料烟品种作为供试材料，对Bagsam和Sanguinetti两种银染方法，在标记检测效率、成本及染色效果 等方面进行了研究，并对其在SRAP标记和TRAP标记中的应用进行了探讨。结果表明，Sanguinetti银染法比Bagsam银染 更为简便、经济，染色照片的色阶图说明Sanguinetti染色方法的背景与扩增带区分明显，能够清楚地读带；且该方法能够 扩增出很好的SRAP和TRAP标记谱带。因此，推荐在SRAP和TRAP标记检测中采用Sanguinetti银染方法。 关键词： 银染 SRAP标记 TRAP标记 Comparison Study of Bassam and Sanguinetti Silver Staining in the Detecting of SRAP and TRAP Ren Min Jia Xinghua Jiang Caihong Yang Aiguo Wang RJxin ( 6∞co Research Institute，Chinese Academy of Agricuhural Sciences，Qingdao 266101) Abstract： The silver staining as a kind of DNA staining methods is important for molecular marker detecting．In order to evaluate the application of the Bagsam and the Sanguinetti silver staining methods in the novelmolecular markersSRAP and TRA P which have been widely applied in many fieldsrecently，two kinds of silver staining were compared in the detecting egiciency，e~en and the images of silver staining with levels by Photoshop computer software．The application in SRA P and TRAP Wag also studied．The results indicated that the Sanguinetti silver staining method Wag simpler，more economy，and easier to distinguish the bandsof PCR amphcation from background than the Bagsam method．It also showed that the Sanguinetti silver staining method had a very good effect on SRAP and TRA P molecular markers． Key words： Silver staining Sequence—R．elated amphfied polymorphism Target region amphfied poly
