　　三磷酸腺苷对大鼠移植胰腺细胞死亡方式的影响 【摘要】 目的 研究三磷酸腺苷(ATP)对大鼠移植胰腺细胞死亡方式的影响，探讨ATP对大鼠移植胰腺缺血再灌注损伤的作用及相关机制。方法 SD大鼠102只，随机分为假手术组、单纯移植组和ATP保护组，后2组按再灌注时间再各分为0、1、6、12 h亚组，单纯移植组供体胰腺单纯用肝素平衡盐液进行低温(0～4℃)灌注并保存60 min后行移植手术，ATP保护组供体胰腺用添加了ATP的肝素平衡盐液进行低温(0～4℃)灌注并保存60 min后行移植手术。观察ATP对血糖、血清脂肪酶、淀粉酶含量和胰腺组织病理学变化的影响，研究胰腺组织ATP含量与凋亡、胀亡细胞百分比的关系。结果 ATP保护组胰腺组织病理损伤明显轻于单纯移植组，再灌注6、12 h ATP保护组血糖、血清淀粉酶、脂肪酶含量明显低于同时间点单纯移植组 (血糖、脂肪酶，Plt;0.05；淀粉酶，Plt;0. 01)，再灌注0、1 h ATP保护组胰腺组织ATP含量明显高于同时间点单纯移植组（Plt;0.01），ATP保护组胀亡细胞百分比明显小于同时间点单纯移植组(0、1、6 h，Plt;0.01；12 h, Plt;0.05)，ATP保护组细胞的主要死亡方式是凋亡，ATP含量与胀亡细胞百分比明显负相关（r=-0.956, Plt;0.01）,与凋亡细胞百分比无明显相关。结论 ATP促进移植胰腺的不可逆损伤细胞以凋亡方式死亡，减少细胞胀亡的发生。 【关键词】 胰十二指肠移植；三磷酸腺苷；细胞凋亡；细胞胀亡 Abstract：Objective To study the effects of adenosine triphosphate (ATP) on the death pattern of transplanted pancreatic cells in diabetic rat models to explore the protective mechanism of ATP against the I/R injury of the graft.Methods 102 drug-induced diabetic SD rats ized into 3 groups: sham operation group, simple transplant group and ATP protection group. The latter te points of reperfusion respectivly. The isolated pancreas from donor rat in before transplantation. ATP as an additive agent ple transplant group. Blood sugar，plasma amylase and lipase concentrations, and ATP content in pancreatic tissue, ined histopathological study later. The percentages of normal, apoptotic, oncotic and necrotic cells ined by floetry.Results It orphological changes of pancreas in ATP group uch milder than those in simple transplant group. The blood sugar，plasma concentrations of amylase and lipase ple transplant subgroups (Plt;0.05, Plt;0.01). The tissue concentration of ATP ple transplant group (Plt;0. 01). The percentage of oncotic cells ple transplant group (Plt;0.01, Plt;0.05); the dead cells ainly apoptotic ones in ATP group.
