大熊猫酸性核糖体磷酸蛋白P1基因cDNA克隆及序列分析.PDF

兽类学报,2008,28 (1):75 - 80 Acta Theriologica Sinica 大熊猫酸性核糖体磷酸蛋白 P1 基因 cDNA 克隆及序列分析  杜玉杰 侯万儒 彭正松 周材权 (西华师范大学生命科学学院, 南充 637002 ) 摘要:运用 RT-PCR 技术,从大熊猫的肌肉组织总 RNA 中成功克隆了酸性核糖体磷酸蛋白P1 (RPLP1 )基因的 表达序列,并对其进行了测序及初步分析。结果表明:大熊猫RPLP1 基因的表达序列全长为448 bp,开放阅读 框 (ORF) 为344 bp,编码114 个氨基酸的蛋白质,该蛋白的分子量为 11 566 kDa,pI 为4 4 ,含有3 个酪蛋白 激酶Ⅱ磷酸化位点和2 个 N - 酰基化位点。进一步分析发现,大熊猫RPLP1 基因的表达序列及其编码的氨基酸 序列与已报道的部分哺乳动物具有很高的相似性。 关键词:大熊猫;RT-PCR;RPLP1 ;克隆;序列分析 中图分类号:Q785 文献标识码:A 文章编号:1000 - 1050 (2008 )01 - 0075 - 06 cDNA cloning and sequences analysis of acidic ribosomal phosphoprotein P1 (RPLP1)from giant panda  DU Yujie,HOU Wanru ,PENG Zhengsong,ZHOU Caiquan (College of Life Science,China West Normal University,Nanchong 637002, China) Abstract :The cDNA of acidic ribosomal phosphoprotein P1 (RPLP1 )was cloned successfully from the giant panda (Ai- luropoda melanoleuca)using RT-PCR technology,which was also sequenced,analyzed preliminarily The result shows that the cDNA fragment cloned is 449 bp in size,and it contains an open reading frame of 344 bp encoding 114 amino acids The deduced protein sequence shows that the protein is composed of 114 amino acids and its estimated molecular weight is 11 566 kDa with a pI of 4 4 Alignment analysis indicated that the nucleotide sequence and the deduced amino acid se- quence are highly conserved to other five species studied,including Homo sapiens ,Mus musculus,Rattus norvegicus ,Bos Taurus and Sus scrofa The homologies for nucleotide sequences of giant panda PPLP1 to that of these species are 92 4% , 89 8% ,89 0% ,91 3% and 87 5% ,while the homologies for amino acid sequences
