反相高效液相色谱光电二极管阵列检测器法测定苦丁茶中乌索酸含 　【摘要】 目的建立苦丁茶中乌索酸含量的测定 方法 ，同时采用光电二极管阵列检测器检测乌索酸色谱峰的纯度。方法色谱柱为Nucleosil C18柱(4.6 mm×250 mm, 5 μm)，流动相为甲醇水(87∶13)，流速0.8 ml/min，检测波长210 nm，柱温26℃。结果乌索酸在1.636～17.724 μg（r=0.999 8）范围内峰面积与进样量呈良好的线性关系，乌索酸的平均回收率为99.3 % (RSD为1.7 %)。结论 该方法样品处理简单，准确度高，分离效能好，适合于苦丁茶叶中乌索酸含量的测定。 中国编辑。 【关键词】 苦丁茶；乌索酸；高效液相色谱；光电二极管阵列检测器 　　Determination of Ursolic Acid in Ilex kudincha by RPHPLCPAD 　　ZOU Shengqin, CHEN Wu*,WU Xiaochun 　　Bioengineering Research Institute of Yichun University·Key Laboratory of Province for Research on Active Ingredients in Natural Medicines,Yichun,336000,China 　　Abstract：ObjectiveTo develop a method for the quantitative analysis of ursolic acid and oleanolic acid in Ilex kudincha C. J. Tseng and at the same time the purity of ursolic acid peak was detected by means of Photodiode Array Detector(PAD).MethodsHPLC was carried out on the Nucleosil C18 column (4.6 mm×250 mm, 5 μm) at 26℃ with MeOHH2O(87∶13) as mobile phase. The flow rate was 0.8 ml and the detection wavelength 210 nm. ResultsThere was good linear relationship between the peak area and the sample content injected in the ranges of 1.636～17.724 μg（r=0.999 8）for ursolic acid. The average recovery rate of ursolic acid was 99.3% (RSD1.7 %). ConclusionThe method is simple, accurate, and suitable for the determination of ursolic acid in Ilex Kudincha C. J. Tseng为冬青科(Aquifoliaceae) 　　冬青属植物，性凉，味甘苦，有散风热清头目、除烦渴的功效。苦丁茶主要分布在广东、广西、湖南、湖北、海南等地，是我国南部及西南部民间传统的天然药用植物，具有清热解毒、生津、提神、益智、抗衰老、调节血压、降脂等药理作用［1］。苦丁茶主要含三萜类、黄酮类、木脂素、氰基化合物、甾醇、氨基酸和苦味素等化学成分［2］。 现代 药理 研究 表明，苦丁茶中的乌索酸具有抗肝炎、抗肿瘤、抗病毒、抗炎抑菌及降血脂血糖等多种药理活性，具有重要的药用开发价值［3］。苦丁茶药理、化学成分研究已有很多报道，但其质量评价方法报道较少。本实验采用高效液相色谱法对不同产地苦丁茶产品中的乌索酸含量进行了测定，为评价其质量提供依据。 　　1 仪器与试药 　　1.1 仪器 　　高效液相色谱仪，Waters515型泵，2996光电二极管阵列检测器，7725I 进样阀，Empower 中文色谱工作站（美国）；RO-MB-10D高纯水机（杭州永洁达膜分离设备厂）；CP2250 分析 天平（德国Sartorius公司）；FW100型高速万能粉碎机（天津市泰斯特仪器有限公司）；KS-1500超声提取
