烫伤大鼠组织脂多糖结合蛋白mrna表达的动态变化（Dynamic changes of lipopolysaccharide binding protein mRNA expression in scalded rats） Dynamic changes of lipopolysaccharide binding protein mRNA expression in scalded rats Update Date: 2010-07-03, Zhai Hongxia, Yao Yongming, Fang Wenhui, Lu Lianrong, Tian Huimin, Sheng Zhiyong [Abstract] Objective To investigate the dynamic expression of lipopolysaccharide binding protein (LBP) mRNA in different tissues after scalding and its relationship with endotoxemia. Methods 35% rats were divided into normal control group, scald group and polymyxin B treatment group. Plasma endotoxin levels were measured by improved pretreatment with perchloric acid, plasma, azo dye, Limulus amebocyte lysate, and tissue LBP and mRNA were determined by RT-PCR method. Results the levels of endotoxin in the portal vein and systemic circulation increased significantly after scalding, and reached the peak at 8 hours. Treatment with polymyxin B may reduce endotoxin levels to varying degrees. Normal tissue can express a small amount of LBP, and after 2 hours scald, the expression of LBP and mRNA in different tissues increased significantly (P0.05 ~ 0.01), and reached peak value at 8 hours, which was about 2 to 2.5 times of the control value, and lasted for 24 hours after injury. Anti endotoxin treatment with polymyxin B could inhibit the expression of LBP and mRNA in lung, intestine and kidney to some extent (P0.05 ~ 0.01). Correlation analysis showed that endotoxin levels in portal vein were positively correlated with LBP and mRNA in lung tissues (r=0.412, P0.05), but there was no significant correlation between endotoxin levels and liver, intestinal and renal tissues. Conclusion the expression of LBP and mRNA in tissue increased significantly after burn. Intestinal endotoxemia might be one of the important factors to stimulate the expression of LBP gene in tissues.
