基质干细胞及Schwann细胞联合移植治疗大鼠 脊髓损伤后后肢功能 　 作者：陈冰，张振兴，王道奎，张强，王玉亭，王金鹏 【摘要】 目的 探讨基质干细胞（mesenchymal stem cells，MSCs）与Schwann细胞（Schwann cells，SCs）联合移植对大鼠脊髓损伤后后肢功能恢复的影响。方法 体外分离纯化大鼠骨髓基质干细胞和Schwann细胞，取SD大鼠30只随机分为对照组、局部移植组和局部、静脉联合移植组，在手术显微镜下制作脊髓横断模型，局部移植组只在损伤局部注射MSCs与Schwann细胞，局部、静脉联合组在局部注射后24 h内经尾静脉注射MSCs与Schwann细胞，对照组在损伤局部注射PBS液。分别于术后7天、14天、21天、28天、35天、42天通过脊髓运动功能评分（BBB评分）观察大鼠后肢功能的恢复情况。结果 局部移植组和局部、静脉联合移植组的评分均显著高于对照组（Plt;005），但局部移植组与联合移植组间无显著差异（Pgt;005）。结论 基质干细胞与Schwann细胞联合移植能有助于脊髓损伤（spinal cord injury，SCI）的恢复。 【关键词】 基质干细胞；许旺细胞；移植；脊髓损伤；大鼠 ［Abstract］ Objective To explore the effects of both MSCs and SCs co-transplantation on the function recovery in rats hind legs.Methods Separating and purifying MSCs and SCs of rats extracorporeally.Taking 30 rats and randomly divided them into a control group，a partially-transplanted group and a co-transplanted group of part and vein.And then made models of crossed spinal cord under the operation microscope.The partially-transplanted group only injected MSCs and SCs in the injured parts.The co-transplanted group of part and vein injected MSCs and SCs through end intravenous injection after the partial injection within 24 hours.The control group injected PBS in the injured parts.Observed the function recovery in rats hind legs through the marks marked by BBB in 7 d，14 d，21 d，28 d，35 d，42 d separately.Results Both of the marks of the partially-injected group and the co-transplantation group of part and vein were much higher than that of the control group （Plt;005）.But the partially-injected group had no clear differences from the co-transplantation group of part and vein （Pgt;005）.Conclusion MSCs and SCs co-transplantation can help the recovery of the SCI. ［Key words］ mesenchymal stem cells;Schwann cell;transplantation;spinal cord injury;rats 实验将大鼠基质干细胞（MSCs）和Schwann细胞（SCs）联合移植于大鼠脊髓横断损伤处及通过静脉移植，观察大鼠后肢功能的恢复，从而反映其对脊髓损伤（spinal cord injury）的修复作用，为临床提供参考。实验在2007年7~11月完成。 1 材料与方法 11 材料 取雌性SD大鼠30只，体重（250±20）g，由解放军第89医院动物实验室提供。MSCs及Schwann细胞由潍坊
