Quarantine inspection and proliferation risks of Japanese

Buhrer) Nikle) associated with other parasites and microbes .

Abstract: To reveal the characteristic and the spread risk of Japanese five-needle pine infected by
pine wilt nematode, the xylem of dead Japanese five-needle pine were collected through the
nursery sampling and the nematode associated with other parasites and microbes were isolated
from chips of the xylem soaking in water; the isolates were inspected under microscope; and then
the larvae (roundhead wood borer) in the xylem of dead Japanese five-needle Pine were further
identified under dissected microscope. When the larvae metamorphose into the beetle of longhorn,
the species of the longhorn beetle were further identified and then the beetle was cut into piece
which was inspected under microscope to find whether or what nematode was carried by the
beetle. The result showed that 33% of the samples were infected by Bursaphelenchus xylophilus;
and 33% of the samples were infected by B. mucronatus ; 66.7%, 83.3% and 100% of the samples
were infested by other invasive protozoon, fungus and bacterium respectively. The larvae
(roundhead wood borer) are the larvae of Monochumus alternatus , which are able to
metamorphose into the beetle of Monochumus alternatus which carry the nematode that is
dauerlarvae LIV of Bursaphelenchus xylophilus. The conclusion is that the
