1小鼠卵母细胞玻璃化冷冻的研究 朱孝荣 孙海燕 孙 强 李厚达 扬州大学比较医学中心 扬州 225009 摘要 目的 对小鼠卵母细胞玻璃化的方法和冷冻液进行研究 方法 用两种玻璃化冷冻 液EFS40 高钠 DEFS40 低钠 对ICR C57BL/6J 小鼠卵母细胞进行冷冻 0.5mol/L 蔗 糖液解冻 体外受精前对部分解冻后的C57BL/6J 小鼠卵母细胞进行透明带切割 结果 DEFS40 冷冻液冷冻效果明显高于 EFS40 冷冻液 ICR 小鼠卵母细胞解冻后成活率达 75.2% 体外 受精率 24.6% 与对照组无明显差异 C57BL/6J 小鼠卵母细胞解冻后成活率为 87.1% 卵 母细胞切割后体外受精率 36.0% 与对照组也无明显差异 结论 冷冻液中钠离子浓度影响 小鼠卵母细胞冷冻效果 用 DEFS40 低钠 冷冻液进行玻璃化冷冻可以取得理想的冷冻效 果 关键词 小鼠 卵母细胞 冷冻 中图分类号 Q782 Study on Vitrifaction of Mouse Oocytes ZHU Xiao-rong,SUN Hai-yan,SUN Qiang,LI Huo-da (Comparative Medicine Center,Yangzhou University,Yangzhou 225009,China) Abstract Objective To explore vitrified method and cryopreservative solution of mouse oocytes. Method The oocytes of ICR and C57BL/6J mice were vitrified with EFS40(high concentration of sodium) and DEFS40(low concentration of sodium),thawed with 0.5mol/L sugar solution.Before fertilization in vitro, zona-pellucieda of C BL/6J mice oocytes 57 were dissected after thawed. Result Considering the vitrified effect , DEFS40 is better than EFS40. The oocyte survival rate of ICR mice reaches 75.2% after thawed . The fertilization rate reaches 24.6% and the difference is not remarkable compared to the control.But the oocyte survival rate of C57BL/6J mice reaches 87.1% after thawed. The oocyte fertilized rate which zona-pellucieda were dissected reaches 46.0% and the difference is not remarkable compared to the control.
